08-P022 Protein phosphatase 2A regulates self-renewal of Drosophila neural stem cells
نویسندگان
چکیده
with ASPM siRNA, there is defective cleavage furrow orientation, cytokinesis failure and an increase in apoptosis. Dominant negative expression of GFP-ASPM C-terminal fusion proteins induces cytokinesis failure in HeLa cells. To further investigate the functions of ASPM in cell division we are currently performing a foetal brain yeast two-hybrid (Y2H) screen to identify direct interactants of the ASPM C-terminal domain. At present, the only reported interactant of the ASPM C-terminus is citron kinase, a protein with a known role in cytokinesis. We will discuss our findings of this association and the results of our Y2H screen.

Protein phosphatase 2A regulates self-renewal of Drosophila neural stem cells.
Drosophila larval brain neural stem cells, also known as neuroblasts, divide asymmetrically to generate a self-renewing neuroblast and a ganglion mother cell (GMC) that divides terminally to produce two differentiated neurons or glia. Failure of asymmetric cell division can result in hyperproliferation of neuroblasts, a phenotype resembling brain tumors. Here we have identified Drosophila Prote...

Asymmetric cell division is a mechanism for generating cell diversity as well as maintaining stem cell homeostasis in both Drosophila and mammals. In Drosophila, larval neuroblasts are stem cell-like progenitors that divide asymmetrically to generate neurons of the adult brain. Mitotic neuroblasts localize atypical protein kinase C (aPKC) to their apical cortex. Cortical aPKC excludes cortical ...

Purpose: Isolation, induction of neural and glial differentiation and evaluating the expression of Nucleostemin, ZFX, Hoxb-4, Sox-9 & Bmi-1 self renewal genes in adult mouse neural stem cells. Materials and Methods: Breifly, for isolation of neural stem cells, frontal part of adult mouse brain was minced in PBS and digested by enzyme solution, containing hyaloronidase and trypsin. Isolated cel...
متن کاملDronc caspase exerts a non-apoptotic function to restrain phospho-Numb-induced ectopic neuroblast formation in Drosophila.
Drosophila neuroblasts have served as a model to understand how the balance of stem cell self-renewal versus differentiation is achieved. Drosophila Numb protein regulates this process through its preferential segregation into the differentiating daughter cell. How Numb restricts the proliferation and self-renewal potentials of the recipient cell remains enigmatic. Here, we show that phosphoryl...
متن کاملProspero acts as a binary switch between self-renewal and differentiation in Drosophila neural stem cells.
Stem cells have the remarkable ability to give rise to both self-renewing and differentiating daughter cells. Drosophila neural stem cells segregate cell-fate determinants from the self-renewing cell to the differentiating daughter at each division. Here, we show that one such determinant, the homeodomain transcription factor Prospero, regulates the choice between stem cell self-renewal and dif...
